The present invention provides an immobilization medium which can immobilize bioreagents to support materials and which dries to a water resistant layer or film. The immobilization medium comprises (i) a liquid or fluid binding reagent and (ii) complexes of a biore- agent immobilized to a solid phase which are evenly dispensed within the binding reagent. The suspension can further include supplemental ingredients evenly dis- persed throughout the medium which can provide the medium with electrochemical properties, enhance the stability of the immobilized bioreagent and/or improve the medium's capability of drying to a substantially water resistant or insoluble layer. The immobilization medium provided by the instant invention is in the form of a homogeneous liquid suspension. The immobiliz- ing medium of the present invention can be employed in essentially any assay format which utilizes an immobi- lized bioreagent. For example, using the immobilization medium, an enzyme electrode can be manufactured and used in conjunction with a counter and reference elec- trode to electrochemically detect the bioconversion of the enzyme's substrate, a biosensor can be manufactured which is capable, via non-electrochemical means, of de- tecting the bioconversion of the enzyme's substrate; and a solid phase can be manufactured which can be used in heterogeneous immunoassay formats known in the art.
La présente invention porte sur un milieu capable d'immobiliser des bioréacteurs pour servir de support à des substances, et qui sèche en formant une couche ou pellicule résistante à l'eau. Ce milieu d'immobilisation comprend (i) un réactif liant le liquide ou le fluide et (ii) des complexes d'un bioréactif immobilisé en phase solide, uniformément dispersés dans le réactif liant. La suspension peut aussi comporter des ingrédients supplémentaires uniformément dispersés dans l'ensemble du milieu, ce qui peut conférer à ce dernier des propriétés électrochimiques, améliorer la stabilité du bioréactif immobilisé et/ou améliorer la capacité du milieu de sécher en formant une couche résistante à l'eau ou insoluble. Le milieu d'immobilisation fourni par la présente invention se présente sous forme de suspension homogène en milieu liquide. Ledit milieu d'immobilisation peut être utilisé dans pratiquement tout protocole de dosage faisant intervenir un bioréactif immobilisé. On peut utiliser ce milieu d'immobilisation, par exemple, pour fabriquer et utiliser un électrode enzymatique en combinaison avec une contre-électrode ou une électrode de référence, afin de détecter électrochimiquement la bioconversion du substrat enzymatique; pour fabriquer un biodétecteur capable de détecter par des moyens non électrochimiques la bioconversion du substrat enzymatique; et pour fabriquer une phase solide susceptible d'être utilisée dans les différents dosages immunologiques connus de la technique actuelle.
