The present invention relates to a human bone morphogenetic protein-4 (BMP-4) promoter region and fragments thereof, a vector comprising said promoter region and host cells comprising said promoter regions or vectors. The invention further relates to a method for the identification of therapeutic agents for use in the prevention and/or treatment of osteoporosis, wherein said method comprises the steps of: a) introducing into a cell a first expression vector comprising a BMP-4 promoter region or fragments thereof operably linked to a reporter gene, and a second expression vector comprising DNA encoding the estrogen receptor; b) contacting said cell with potential therapeutic agents; c) monitoring the expression of the protein encoded by the reported gene.
La présente invention concerne une région promotrice de la protéine morphogénétique osseuse 4 (BMP-4) chez l'humain et des fragments de ladite région, un vecteur comprenant ladite région promotrice, et des cellules hôtes comprenant lesdites régions promotrices ou vecteurs. L'invention concerne en outre un procédé qui permet d'identifier des agents thérapeutiques pouvant être utilisés pour la prévention et/ou le traitement de l'ostéoporose. Ledit procédé consiste à a) introduire à l'intérieur d'une cellule un premier vecteur d'expression comprenant une région promotrice de la BMP-4 (ou des fragments de ladite région) liée de manière fonctionnelle à un gène marqueur, et un deuxième vecteur d'expression comprenant l'ADN codant le récepteur des oestrogènes; b) mettre ladite cellule en contact avec des agents thérapeutiques potentiels; c) contrôler l'expression de la protéine codée par le gène marqué.
