This invention pertains to BIV constructs encompassing BIV combination vectors, BIV vectors and BIV packaging vectors and particularly the invention pertains to a three vector system comprising: a) a BIV vector construct including a DNA segment from a BIV genome, a packaging sequence to package RNA into virions; a promoter operably linked to the DNA segment; and a transgene operably linked to a second promoter; b) a BIV packaging vector construct comprising a BIV DNA sequence fragment comprising at least a gag gene or pol gene of BIV; a promoter operably linked to the BIV DNA fragment; and a polyadenylation sequence located downstream of the BIV DNA fragment; and c) an expression vector construct comprising a gene encoding a viral surface protein. Also provided is a method for transferring a gene of interest into a mammalian cell.
L'invention concerne des produits génétiques de BIV englobant des vecteurs de recombinaison de BIV, des vecteurs de BIV et des vecteurs d'encapsidation de BIV et, en particulier, un système basé sur trois vecteurs et comprenant: (a) un produit génétique de vecteur de BIV comportant un segment d'ADN provenant d'un génome de BIV, une séquence d'encapsidation servant à encapsider ARN dans des virions; un promoteur lié aux segments d'ADN; un transgène lié à un deuxième promoteur; (b) un produit génétique de vecteur d'encapsidation de BIV comprenant un fragment de segment d'ADN de BIV possédant au moins un gène gag ou un gène pol de BIV; un promoteur lié aux fragments d'ADN de BIV; une séquence de polyadénylation située en aval du fragment d'ADN de BIV; (c) un produit génétique de vecteur d'expression comprenant un gène codant une protéine de surface virale. Elle concerne également un procédé servant à transférer un gène dans une cellule mammifère.
