Described is a novel gene and its encoded secreted tumor antigen, termed BPC- 1, and to diagnostic and therapeutic methods and compositions useful in the management of various cancers which express BPC-1, particularly including prostate cancer and bladder cancer. In human normal tissues, BPC-1 is only expressed in certain tissues of the brain. However, BPC-1 is expressed at high levels in prostate cancer cells and is also expressed in bladder cancer cells. The structure of BPC-1 includes a signal sequence and a CUB domain. BPC-1 protein is secreted. Preliminary experimental evidence suggests that BPC-1 is directly involved in oncogenesis or maintenance of the transformed phenotype of cancer cells expressing BPC-1. BPC-1 also appears to bind specifically to a cellular protein expressed in prostate cancer cells and other cells.
L'invention concerne un nouveau gène ainsi que l'antigène tumoral de celui-ci, sécrété, codé et dénommé BPC-1; elle concerne également des méthodes et compositions diagnostiques et thérapeutiques, utiles dans la gestion de divers cancers exprimant BPC-1, notamment le cancer de la prostate et celui de la vessie. Dans des tissus normaux humains, BPC-1 n'est exprimé que dans certains tissus du cerveau. Cependant, BPC-1 est exprimé à des niveaux élevés dans les cellules du cancer de la prostate, de même que dans les cellules du cancer de la vessie. La structure de BPC-1 comprend une séquence de signaux ainsi qu'un domaine CUB. La protéine BPC-1 est sécrétée. Une évidence expérimentale préliminaire suggère que BPC-1 est directement impliqué dans l'oncogenèse ou le maintien du phénotype transformé de cellules cancéreuses exprimant BPC-1. BPC-1 semble également se lier spécifiquement à une protéine cellulaire exprimée dans les cellules du cancer de la prostate ainsi que dans d'autres cellules.
