The current invention relates to an active factor present in homogenized brain tissue which inhibits antigen-driven proliferation of lymphocytes in culture, but stimulates proliferation in response to most mitogens. The inhibitory activity can be destroyed by treatment with proteases or neuraminadase. The activity is in the insoluble fraction of the homogenate, but becomes soluble in 0.04 M NaOH. After gel filtration chromatography of the NaOH soluble material, the suppressive activity is in the high molecular weight fraction which elutes in the void volume of the column. This fraction contains protein and carbohydrate. The activity is not affected by neutralizing antibodies against regulatory cytokines, does not depend on Fas or FasL, and is not due to the presence of gangliosides. These data suggest that a brain glycoprotein which is either membrane-bound or part of the extracellular matrix has powerful immune regulatory effects in culture.
L'invention concerne un facteur actif présent dans des tissus cérébraux homogénéisés inhibant la prolifération induite par antigène des lymphocytes dans une culture, mais stimulant la prolifération en réponse à la plupart des mitogènes. Il est possible d'annihiler l'activité inhibitrice par un traitement à base de protéases ou de neuraminadase. L'activité se situe dans la fraction insoluble de l'homogénéisat, mais la solubilité apparaît dans du NaOH concentré à 0,04 M. Après une chromatographie à filtration en milieu gélifié du matériau soluble NaOH, l'activité inhibitrice apparaît dans la fraction à poids moléculaire élevé, ce qui donne une élution dans le volume vide de la colonne. La fraction renferme des protéines et des hydrates de carbone. L'activité n'est pas affectée par la neutralisation des anticorps vis-à-vis des cytokines régulatrices, elle n'est pas Fas ou FasL dépendante, et elle n'est pas imputable à la présence de gangliosides. Cela tend à montrer qu'une glycoprotéine cérébrale, membranaire ou appartenant à la matrice extracellulaire, produit de puissants effets immunorégulateurs en culture.
