The present invention provides methods and kits using low conductivity buffers for the detection of mRNA from a cell lysate separated in a microfluidic device. The buffers do not affect the integrity of the cell nucleus, thereby allowing for generation of a cell lysate that can be directly used as a template in reverse transcriptase (RT)-polymerase chain reaction (PCR) amplification reactions without contaminating genomic DNA.
La présente invention concerne des procédés et des kits utilisant des tampons à faible conductivité destinés à détecter l'ARN messager présent dans un lysat de cellules, répartis dans un dispositif microfluidique. Les tampons n'affectent pas l'intégrité du noyau de cellule, permettant par conséquent la génération d'un lysat de cellules qui peut directement servir de matrice dans des réactions de l'amplification en chaîne par polymérase inverse, sans contaminer l'ADN génomique.
