72222-87 ABSTACT OF THE DISCLOSURE Erythromycin A derivatives of the formula: (3) Image (wherein: R is hydroyen or (C2-C4)alkanoyl; R1 and R2 are taken separately, R1 is hydxoxymethyl and R2 is hydroxy; or R1 and R2 are taken together and are methylene; and R3 and R4 are taken separately and are each hydro- gen; or R3 and R4 are taken together and are thiocarbonyl; with the proviso that when R3 and R4 are so taken, R is other than hydrogen); and their pharmaceutical acceptable acid addition salts are anti- bacterial and useful as active ingredients of pharmaceuticals. These derivatives can be produced by selective formation of the exocylic double bond at the C-12 position (i.e., when R1 and R2 are methylene) from corresponding 12-beta-hydroxy-12-alpha-methyl compounds while the 9 and 11-hydroxyl groups are thiocarbonate, followed by diol formation and/or deacylation as required.
