The invention relates to a precursor of human insulin or an insulin analogue of the formula (I): Fus-B(1-30)-RDVP-Yn-A(1-21), whereby Fus is an optionally present fusion component of any sequence, B(1-30) is the B chain of human insulin, Y stands for an amino acid chain which ends with a C-terminal basic amino acid, n is 2 to 50 and indicates the length of the amino acid chain Y and A(1-21) is the A chain of human insulin. The A and/or the B chain can be modified by means of amino acid exchanges, deletions and/or additions. The invention also relates to a DNA coding therefor. The invention further relates to the production and the use of the precursor and the DNA as well as to a method for producing human insulin or an insulin analogue.
L'invention concerne un précurseur de l'insuline humaine ou d'un analogue de cette dernière, de formule (I) : Fus-B(1-30)-RDVP-Y¿n?-A(1-21) (I), dans laquelle Fus représente un élément de fusion éventuellement présent, de séquence quelconque; B(1-30) représente la chaîne B de l'insuline humaine, Y représente une chaîne d'acides aminés se terminant par un acide aminé basique C-terminal ; n est compris entre 2 et 50, et indique la longueur de la chaîne Y d'acides aminés ; et A(1-21) représente la chaîne A de l'insuline humaine. La chaîne A et/ou la chaîne B peuvent être modifiées par des échanges, des délétions et/ou des additions d'acides aminés. L'invention concerne également un ADN codant pour ce précurseur, la production et l'utilisation de ce précurseur et de cet ADN, ainsi qu'un procédé pour produire de l'insuline humaine ou un analogue de cette dernière.
