Disclosed are polypeptide analogues of human C5a which are C5a receptor antagonists that exhibit substantially no analphylatoxin or agonist activity, and derivatives of the analogues, and dimeric forms of the analogues or derivatives. DNA molecules encoding the polypeptides and methods of making the analogues are also provided. Pharmaceutical formulations containing a C5a analogue are used therapeutically in the treatment of C5a-medicated diseases and inflammatory conditions in mammals, and prophylactically to prevent or reduce inflammation caused by an event which causes inflammation or aggravates an existing inflammatory condition, respectively. Further disclosed are antibodies specific to the C5a analogues, derivatives thereof, and dimers of the analogues and derivatives which exhibit substantially no cross-reactivity with human C5a. The antibodies are used to detect or quantify circulating C5a analogue or derivative, as well as to modify, e.g., neutralize, the activity of the C5a receptor antagonist in vivo.
Cette invention concerne des analogues polypeptidiques de la protéine C5a humaine qui sont des antagonistes du récepteur de C5a ne présentant pratiquement aucune activité d'analphylatoxine ou d'agoniste, et des dérivés de ces analogues, ainsi que des formes dimères de ces analogues ou dérivés. On décrit aussi des molécules d'ADN codant ces polypeptides et des procédés de préparation de ces analogues. On utiilise, à des fins thérapeutiques, des formulations pharmacetiques contenant un analogue de C5a, pour traiter des maladies et des pathologies inflammatoires induites par la C5a chez les mammifères, et à des fins proplylactiques, pour respectivement empêcher ou réduire l'inflammation provoquée par un évènement qui provoque l'inflammation ou aggrave une pathologie inflammatoire existante. On décrit également des anticoprs spécifiques contre les analogues de C5a, des dérivés de ces derniers, ainsi que de ces dimères de ces analogues et dérivés qui ne présentent pratiquement aucune réactivité croisée avec la C5a humaine. On utilise ces anticorps pour détecter ou quantifier l'analogue de C5a ou son dérivé en circulation, ainsi que pour modifier, par exemple neutraliser, l'activité de l'antagoniste du récepteur de C5a in vivo.
