The invention pertains to a protein which comprises the same or substantially the same amino acid sequence as that represented by Fig. 3, or a variant of the amino acid sequence having a deletion, addition or substitution of amino acids, an isolated cDNA comprising DNA having a nucleotide sequence encoding this protein, or a DNA hybridizing with this DNA, or a DNA which is synonymous to the said DNAs, a recombinant vector comprising the DNA. The invention furthermore pertains to methods of screening for and detection of pNEU60 mutation carriers with macular degeneration, prenatal pNEU60 screening and diagnosis, gene therapy utilizing recombinant DNA as well as the generation of transgenic animals (knock-in, knock-out, transgenic animals), anti-pNEU60 antibodies and use in screening for modulating drugs.
L'invention se rapporte à une protéine qui comporte la même séquence ou sensiblement la même séquence d'acides aminés que celle représentée par la figure 3, ou à un variant de cette séquence d'acides aminés présentant une délétion, une addition ou une substitution d'aides aminés, à un ADNc isolé comportant un ADN ayant une séquence nucléotidique codant pour cette protéine, ou un ADN susceptible de s'hybrider avec l'ADN, ou un ADN qui est synonyme des ADN précédents, et à un vecteur recombiné comportant cet ADN. L'invention se rapporte en outre à des procédés de sélection et de détection de porteurs de mutation pNEU60 à dégénérescence maculaire, à la sélection et au diagnostic prénatal de pNEU60, à une thérapie génique utilisant un ADN recombiné ainsi qu'à la génération d'animaux transgéniques (animaux à gène activé ou dits "knock-in", animaux à gène désactivé ou dits "knock-out", animaux transgéniques) et d'anticorps dirigés contre les protéines pNEU60 et à leur utilisation pour la sélection de médicaments modulateurs.
