The present invention provides CDR-grafted antibodies against human tissue factor that retain the high binding affinity of rodent monoclonal antibodies against tissue factor but have reduced immunogenicity. The present humanized antibodies are potent anticoagulants and are thus useful in the treatment and prophylaxis of human thrombotic disease. The invention also provides methods of making the CDR-grafted antibodies and pharmaceutical compositions for the attenuation or prevention of coagulation.

La présente invention concerne des anticorps contre la thromboplastine tissulaire humaine comportant des régions de détermination complémentaire greffées, qui conservent l'affinité de liaison élevée d'anticorps monoclonaux de rongeurs contre la thromboplastine tissulaire mais présentent une immunogénicité réduite. Ces anticorps humanisés constituent de puissants anticoagulants et sont donc utiles pour le traitement et la prophylaxie de thromboses chez l'homme. L'invention concerne également des méthodes de production d'anticorps greffés sur des régions de détermination complémentaire et des compositions pharmaceutiques servant à atténuer et à prévenir la coagulation.
