Methods are taught for culturing mammalian cells, preferably human cells, to improve production of proteins, recombinant or endogenous. Methods are also provided for the growth and long-term survival of cell lines, particularly cell lines established from primary culture. Cell culture media are also provided, contained varying levels of selected amino acids, supplemented with various growth factors and trace elements. In additionally, the media are optionally serum-free, and preferably use an energy source other than glucose. The media are particularly suitable for the primary culture and long-term culture of human fetal cells.
L'invention concerne des procédés permettant de cultiver des cellules mammifères, de préférence, des cellules humaines, aux fins d'amélioration de la production de protéines, recombinantes ou endogènes. L'invention concerne également la croissance et la survie à long terme de lignes cellulaires, notamment celles établies à partir d'une culture primaire. L'invention concerne, en outre, des milieux de culture de cellules comprenant des niveaux variables d'acides aminés sélectionnés, renforcés par divers facteurs de croissance et d'éléments de trace. De plus, les milieux sont éventuellement exempts de sérum et utilisent, de préférence, une source d'énergie autre que le glucose. Les milieux sont spécialement conçus pour la culture primaire et la culture à long terme de cellules de foetus humains.
