2070660 9109114 PCTABS00005 In the field of regulation of cell proliferation, a method of inhibiting or preventing cell proliferation is disclosed. Rb, a stable protein encoded by the retinoblastoma gene (RB1), is synthesized at all phases of the cell cycle. Newly synthesized Rb is underphosphorylated during the G0 and G1 phases but is phosphorylated at multiple sites at the G1/S boundary and during S phase. HL-60 cells lose their ability to multiply phosphorylate newly synthesized Rb when induced to terminally differentiate by chemicals such as dimethylsulfoxide, retinoic acid and butyric acid. A method of using these chemicals to inhibit or prevent cell proliferation in cells containing Rb is disclosed.
