It is intended to provide an extract, a kit and a method for synthesizing a protein in a cell-free system. The above-described extract is characterized by being prepared from mutant Escherichia coli cells having a mutation in an S12 ribosomal protein gene. In a preferred embodiment, the above mutation is such a mutation as imparting tolerance to streptomycin or dependence thereon to E. coli whereby the reading efficiency of mRNA codon on ribosome can be improved and thus the protein productivity can be elevated.

L'invention concerne un extrait, une trousse et un procédé permettant de synthétiser une protéine dans un système acellulaire. Cet extrait est caractérisé en ce qu'il est préparé à partir de cellules mutantes d'Escherichia Coli qui comportent une mutation d'un gène de la protéine ribosomique S12. Dans une forme de réalisation préférée, cette mutation confère à E. coli une résistance ou une dépendance à la streptomycine, ce qui permet d'améliorer l'efficacité de lecture du codon d'ARNm sur le ribosome, et d'augmenter par conséquent la productivité de la protéine.
