An aqueous composition and a method of use are disclosed for the separation of different living cells, such as pancreatic islet and acinar cells, The composition comprises a water soluble, metabolically inert substance to control density, osmolarity and pH; a physiological cold storage solution containing viscosity modifiers to increase the viscosity and osmotically active, impermeant agents to suppress cold-induced edema; all in a medium capable of maintaining viable cells. The solutions are combined to provide a first solution with a density greater than both cell types in their natural state but less than the second cell type after incubation in said solution; and a second solution with a density less than that of both cell types. Separation is achieved by allowing the system to stand or by centrifugation.

L'invention se rapporte à une composition aqueuse et à un procédé d'utilisation d'une telle composition dans la séparation de cellules vivantes différentes, telles que les cellules des îlots du pancréas et les cellules acineuses. La composition comprend une substance hydrosoluble métaboliquement inerte, qui sert à réguler la densité, l'osmolarité et le pH; une solution physiologique de stockage à froid contenant des modificateurs de viscosité augmentant la viscosité et des agents d'imperméance à action osmotique détruisant les oedèmes induits par le froid, tous contenus dans un milieu capable de maintenir les cellules viables. Les solutions sont combinées pour former une première solution ayant une densité supérieure à celle des deux types de cellules à leur état naturel mais inférieure à celle du second type de cellules après incubation dans cette solution, ainsi qu'une seconde solution ayant une densité inférieure à celle des deux types de cellules. On obtient la séparation en laissant reposer le système ou en effectuant une centrifugation.
