The present invention relates to cells for the production of helper dependent adenoviral vectors, including at least the following genie units: a first genie unit comprising an adenovirus defective genome having the inverted terminal repeats in head-to-tail configuration, the encapsidation signal inactivated, and at least one of the non-structural regions inactivated; a second genie units comprising at least one inducible promoter and at least one of the regions inactivated in the first genie unit, said regions being under the control of said inducible promoter; whereby following the activation of the inducible promoter of the second genie unit and the infection of the cells with said helper dependent adenoviral vectors, the first genie unit and the second genie unit enable the production of said helper dependent defective adenoviral in said cells in absence of helper.
La présente invention concerne des cellules pouvant être utilisées pour produire des vecteurs adénoviraux dépendant de virus auxiliaires en l'absence d'autres virus auxiliaires, ces cellules contenant des motifs géniques capable d'exprimer les fonctions nécessaires à l'achèvement d'un cycle viral, fonctions qui sont absentes des vecteurs susmentionnés. La présente invention concerne également un procédé de production de ces cellules, ainsi que leur utilisation pour produire des vecteurs adénoviraux.
