The present invention concerns compositions, methods and/or apparatus of central administration of various CNS-active agents. In particular embodiments, intrathecal administration is advantageous for decreasing the systemic concentrations of CNS agent, thereby decreasing side effect toxicity, while allowing more effective delivery of the agent to the site of action, simultaneously decreasing the dosage delivered to the subject. In particular embodiments, ICV delivery may be of use for patients who have previously proven to be refractory to systemic administration of CNS agents, in some cases due to systemic side effects, or for those patients whose symptoms are of sufficient severity to warrant more aggressive therapeutic intervention. ICV administration allows not only lower systemic concentration but also higher therapeutically effective concentration within the CNS.
La présente invention concerne des compositions, des procédés et/ou un appareil pour l'administration par voie centrale de divers agents actifs dans le SNC. Dans des modes de réalisation particuliers, l'administration par voie intrathécale est avantageuse pour abaisser les concentrations systémiques de l'agent SNC, et diminuer ainsi la toxicité des effets secondaires, tout en permettant une délivrance plus efficace de l'agent sur le site d'action, et une réduction simultanée de la dose délivrée au sujet. Dans des modes de réalisation particuliers, une délivrance par ICV peut être utile chez les patients qui se sont précédemment avérés réfractaires à l'administration par voie systémique d'agents SNC, dans certains cas, en raison des effets secondaires systémiques, ou chez les patients dont les symptômes sont d'une sévérité suffisante pour justifier une intervention thérapeutique plus agressive. L'administration par ICV permet non seulement une concentration systémique plus basse mais aussi une concentration thérapeutiquement efficace plus élevée dans le SNC.
