Provided is a method for characterising a polypeptide or a population of polypeptides, which method comprises the steps of: (a) contacting a sample comprising one or more polypeptides with a lysine reactive agent to cap .epsilon.-amino groups; (b) optionally reacting the sample of polypeptides with an amine reactive reagent to block .alpha.-amino groups; (c) digesting the sample of polypeptides with a cleavage reagent to produce peptide fragments; (d) optionally deactivating the cleavage reagent; (e) removing those peptides having uncapped or unblocked amino groups; and (f) recovering the N-terminal peptides.

Procédé visant à caractériser un polypeptide ou une population de polypeptides. Ce procédé comporte les étapes consistant à : (a) mettre en contact un échantillon contenant un ou plusieurs polypeptides avec un agent réactif à sélectivité lysine pour coiffer les groupes amino .epsilon. ; (b) faire éventuellement réagir l'échantillon de polypeptides avec un réactif sensible aux amines pour bloquer les groupes amino .alpha. ; (c) faire digérer l'échantillon de polypeptides par un réactif de coupure afin d'obtenir des fragments de peptides ; (d) éventuellement désactiver le réactif de coupure ; (e) éliminer les peptides contenant des groupes amino sans coiffe ou débloqués ; et (f) extraire les peptides N-terminaux.
