Provided is a method for characterising a polypeptide or a population of polypeptides, which method comprises: (a) contacting a sample comprising one or more polypeptides with a first cleavage agent to generate polypeptide fragments; (b) isolating one or more polypeptides fragments, each fragment comprising the N-terminus or the C- terminus of the polypeptide from which it was fragmented; (c) identifying the isolated fragments by mass spectrometry; (d) repeating steps (a)-(c) on the sample using a second cleavage agent that cleaves at a different site from the first cleavage agent; and (e) characterising the one or more polypeptides in the sample from the fragments identified in steps (c) and (d).
L'invention concerne un procédé de caractérisation d'un polypeptide ou d'une population de polypeptides, qui consiste à: (a) mettre un échantillon contenant un ou plusieurs polypeptides en contact avec un premier agent de clivage en vue de générer des fragments de polypeptides; (b) isoler un ou plusieurs fragments de polypeptides, comprenant chacun la terminaison N ou la terminaison C du polypeptide dont il est tiré; (c) identifier les fragments isolés par spectrométrie de masse; (d) répéter les étapes (a)-(c) sur l'échantillon au moyen d'un second agent de clivage qui clive sur un site différent de celui qui était utilisé pour le premier clivage; et (e) caractériser le ou les polypeptides dans l'échantillon à partir des fragments identifiés aux étapes (c) et (d).
