Severe Acute Respiratory Syndrome ("SARS") is a human respiratory disease of recent origin, widespread infectivity, recurring incidence, and significant mortality. Although there is abundant evidence suggesting that the coronavirus responsible for the disease ("SARS-CoV") evolves during an outbreak, there is currently little data on the earliest strains of this coronavirus. The present invention is directed to the characterization of the genomic RNA sequences of these earliest SARS coronaviruses, to the identification of nucleotide positions within the SARS-CoV genomic RNA that are characteristic of the different evolutionary stages of the coronavirus, to kits based on these positions for use in diagnosis of the disease in patients, and for the development of vaccines to the disease based on the lowered virulence and contagiousness of these earliest strains of SARS-CoV.
Le syndrome respiratoire aigu sévère (SRAS) est une maladie respiratoire chez l'homme, qui est d'origine récente, présente un pouvoir infectieux important, est récurrente et a un taux de mortalité considérable. Bien qu'il existe de nombreuses indications qui suggèrent que le coronavirus à l'origine de la maladie (le <= SRAS-CoV >=) subit une évolution au cours de chaque flambée, il n'existe que peu d'informations concernant les souches les plus précoces de ce coronavirus. La présente invention se rapporte à la caractérisation des séquences d'ARN génomique de ces coronavirus de SRAS les plus précoces, à l'identification des positions nucléotidiques au sein de l'ARN génomique du SRAS-CoV qui caractérisent les différents stades évolutifs de ce coronavirus, et à des trousses à base de ces positions, utilisables dans le diagnostic de la maladie chez les malades, et dans l'élaboration de vaccins dirigés contre ladite maladie, sur la base de la virulence et la contagiosité réduites de ces souches les plus précoces du SRAS-CoV.
