Disclosed is a chemically inducible promoter for transforming plants or plant cells with genes which are regulatable by adding the plants or cells to a medium containing an inducer or by removing them from such medium. The promoter is inducible by a glucocorticoid, estrogen or inducer not endogenous to plants. Such promoters may be used with any plant genes that can promote shoot regeneration and development to induce shoot formation in the presence of a glucocorticoid, estrogen or inducer. The promoter may be used with antibiotic or herbicide resistance genes or other genes which are regulatable by the presence or absence of a given inducer. Also presented are organisms or cells comprising a gene wherein the natural promoter of the gene is disrupted and the gene is placed under the control of a transgenic inducible promoter. These organisms and cells and their progeny are useful for screening for conditional gain of function and loss of function mutations.

L'invention concerne un promoteur que l'on peut induire chimiquement pour transformer des plantes ou des cellules v~g~tales au moyen de g­nes que l'on peut r~guler en ajoutant des plantes ou des cellules ~ un milieu contenant un inducteur ou en les ~liminant de ce milieu. On peut induire le promoteur au moyen d'un glucocortico~de, un oestrog­ne ou un inducteur non endog­ne chez les plantes. On peut utiliser de tels promoteurs avec un quelconque g­ne v~g~tal capable de favoriser la r~g~n~ration et le d~veloppement de pousses pour induire une formation de pousses en pr~sence d'un glucocortico~de, d'un oestrog­ne ou d'un inducteur. On peut utiliser le promoteur avec des g­nes de r~sistance aux antibiotiques ou aux herbicides ou d'autres g­nes qui peuvent Útre r~gul~s par la pr~sence ou l'absence d'un inducteur donn~. L'invention concerne ~galement des organismes ou des cellules comportant un g­ne dans lequel le promoteur naturel du g­ne est rompu, le g­ne ~tant alors plac~ sous la r~gulation d'un promoteur inductible transg~nique. Ces organismes et cellules ainsi que leur descendance sont utiles pour rechercher des mutations ~ gain et ~ perte de fonctions.
