The invention relates to an HIV envelope glycoprotein in a purified form which can be obtained using a method comprising the following steps: (1) obtaining an envelope glycoprotein in a purified form; (2) reducing at least one disulphide bond of the protein from step (1); (3) alkylating at least two free sulfhydryl groups; (4) optionally, oxidizing the remaining sulfhydryl group; (5) denaturing and (6) renaturing. The invention also relates to the use of the inventive modified HIV envelope glycoprotein in a vaccine against HIV which can be used to induce HIV-neutralising antibodies in a human subject for therapeutic or prophylactic purposes.
La présente invention a pour objet une glycoprotéine d'enveloppe du VIH sous forme purifiée susceptible d'être obtenue par un procédé comprenant les étapes suivants: (1) obtention d'une glycoprotéine d'enveloppe sous forme purifiée, (2) réduction d'au moins un pont disulfure de la protéine de l'étape (1), (3) alkylation d'au moins deux groupes sulfhydryls libres, (4) éventuellement oxydation des groupes sulfhydryls libres restants, (5) dénaturation et (6) renaturation et son utilisation dans un vaccin contre le VIH utilisable pour induire des anticorps neutralisants le VIH chez un sujet humain à titre thérapeutique ou prophylactique.
