The present invention relates to method for screening chemically modified mutant enzymes for amidase and/or esterase activity. This method includes providing a chemically modified mutant enzyme with one or more amino acid residues from an enzyme being replaced by cysteine residues, where at least some of the cysteine residues are modified by replacing thiol hydrogen in the cysteine residues with a thiol side chain, contacting the chemically modified mutant enzyme with a substrate for an amidase and/or a substrate for an esterase and determining whether the chemically modified mutant enzyme exhibits amidase and/or esterase activity. The presentinvention also relates to chemically modified mutant enzymes and a method for producing them where one or more amino acid residues from an enzyme are replaced by cysteine residues, and the cysteine residues are modified by replacing at least some of the thiol hydrogen in the cysteine residue with a thiol side chain to form the chemically modified mutant enzyme. The thiol side chain is selected from the group consisting of -SCH2(p-CH3-C6H4), -SCH2(p-OCH3-C6H4), -SCH2(p-CF3-C6H4), and -SCH2(2,4-diNO2- C6H3).
L'invention concerne un procédé de criblage d'enzymes mutantes chimiquement modifiées visant l'activité d'amidase et/ou d'estérase. Le procédé consiste à fournir une enzyme mutante chimiquement modifiée, un ou plusieurs résidus d'acides aminés d'une enzyme étant remplacés par des résidus de cystéine, où l'on a modifié au moins certains résidus de cystéine en remplaçant l'hydrogène de thiol dans les résidus de cystéine avec une chaîne latérale de thiol, en mettant l'enzyme mutante chimiquement modifiée en contact avec un substrat pour une amidase et/ou un substrat pour une estérase et en déterminant si l'enzyme mutante chimiquement modifiée manifeste une activité d'amidase et/ou d'estérase. La présente invention se rapporte également à des enzymes mutantes chimiquement modifiées et à un procédé de leur fabrication dans des conditions où un ou plusieurs résidus d'acides aminés provenant de l'enzyme sont remplacés par des résidus de cystéine et où les résidus de cystéine sont modifiés par le remplacement d'au moins une partie de l'hydrogène de thiol dans le résidu de cystéine avec une chaîne latérale de thiol, et ce pour former l'enzyme mutante chimiquement modifiée. La chaîne latérale de thiol est sélectionnée dans le groupe constitué de -SCH2(p-CH3-C6H4), -SCH2(p-OCH3-C6H4), -SCH2(p-CF3-C6H4) et -SCH2(2,4-diNO2-C6H3).
