ABSTRACT OF THE DISCLOSURE A chemiluminescent method using a protected enhancer compound which is triggered by a hydrolytic enzyme and then enhances a chemiluminescent reaction is described. The reaction involves an amino substituted acylhydrazide which is activated to produce light by a peroxide compound and a peroxidase in the presence of the activated enhancer compound. The result is enhanced generation of light from the reaction. The hydrolytic enzyme is present alone or as a label in an immunoassay, DNA probe assay or other assay wherein the hydrolytic enzyme is bound to a reporter molecule.
