A chimeric adenoviral vector is provided that comprises nucleotide sequence of a first adenovirus, wherein all or part of at least one gene of said first adenovirus encoding a protein that facilitates binding of said vector to a target mammalian cell, or internalization thereof within said cell, is replaced by all or part of the corresponding gene from a second adenovirus belonging to subgroup D, said vector further comprising a transgene operably linked to a eucaryotic promoter to allow for expression therefrom in a mammalian cell. Compositions comprising such vectors and methods of using such vectors to deliver transgenes to target mammalian cells, particularly airway epithelial cells, are also provided.
Vecteurs d'adénovirus chimériques comportant une séquence nucléotidique d'un premier adénovirus, dans lequel la totalité ou une partie d'au moins un gène de ce premier adénovirus, codant une protéine qui facilite la liaison de ce vecteur à une cellule cible de mammifère, ou la pénétration du vecteur à l'intérieur de la cellule, est remplacée par la totalité ou une partie du gène correspondant d'un second adénovirus appartenant au sous-groupe D. Ce vecteur comporte en outre un transgène coopérant avec un promoteur eucaryote pour promouvoir l'expression du vecteur dans une cellule de mammifère. L'invention porte également sur des compositions contenant ces vecteurs et sur des procédés utilisant ces vecteurs pour transporter des transgènes dans des cellules cibles de mammifères, en particulier dans les cellules épithéliales des voies respiratoires.
