Antibody molecules specific for surface structures of antigen presenting cells that have been modified to include an antigen moiety at a specific site therein to produce novel conjugate antibody molecules are disclosed. These conjugate molecules are produced by genetic modification of genes encoding light and heavy chains of the surface structure specific antibody, and expression in mammalian cells to produce the conjugate antibody. The conjugate antibody retained specificity for antigen presenting cells and contained the antigen moiety. The conjugate antibody molecules deliver the antigen to antigen presenting cells to produce an enhanced immune response to a host immunized therewith. The conjugate antibody molecules and nucleic acid molecules encoding them are useful as antigens and as immunogens in diagnostic and prophylactic applications.
L'invention concerne des molécules d'anticorps spécifiques aux structures de la surface des cellules présentatrices de l'antigène. Ces molécules ont été modifiées pour contenir une fraction antigène à un site spécifique, formant ainsi des molécules nouvelles d'anticorps conjugué. On produit ces molécules conjuguées en modifiant génétiquement les gènes codant les chaînes légères et lourdes de l'anticorps spécifique à la structure cellulaire superficielle et en les exprimant dans des cellules de mammifères afin de produire l'anticorps conjugué. L'anticorps conjugué garde sa spécificité par rapport aux cellules présentatrices de l'antigène et contient la fraction antigène. Les molécules d'anticorps conjugué acheminent l'antigène aux cellules présentatrices de l'antigène, déclenchant une réponse immunitaire améliorée chez l'hôte immunisé avec ces molécules. Les molécules d'anticorps conjugué et les molécules d'acide nucléique qui les codent sont utiles comme antigènes et immunogènes pour des applications diagnostiques et prophylactiques.
