Chimeric complement inhibitor proteins are provided which include a first functional domain (first amino acid sequence) having C3 inhibitory activity and a second functional domain (second amino acid sequence) having C5b-9 inhibitory activity. The first functional domain is amino terminal to the second functional domain. In this way, the chimeric protein exhibits both C3 and C5b-9 inhibitory activity. The other orientation i.e., the orientation in which the second amino acid sequence is amino terminal to the first amino acid sequence, only produces C3 inhibitory activity. Nucleic acid molecules encoding such proteins are also provided.
L'invention concerne des protéines chimères inhibitrices de complément comprenant un premier domaine fonctionnel (première séquence d'acides aminés) présentant un activité inhibitrice de C3, et un second domaine fonctionnel (seconde séquence d'acides aminés) présentant une activité inhibitrice de C5b-9. Le premier domaine fonctionnel est amino-terminal par rapport au second domaine fonctionnel. Ainsi, la protéine chimère présente une activité inhibitrice de C3 et de C5b-9. L'autre orientation, celle dans laquelle la seconde séquence d'acides aminés est amino-terminale par rapport à la première séquence d'acides aminés ne produit qu'une activité inhibitrice de C3. Des molécules d'acide nucléique codant ces protéines sont également décrites.
