A chimeric protein comprises a redox catalytic domain from one source and an electron transfer domain from a different source. The protein is used in a method in which a substrate for the redox catalytic domain is acted on, electrons are transferred between the redox catalytic domain and the electron transfer domain and between the electron transfer domain and an electrode. The flow of current or potential at the electrode may be monitored to determine the presence or amount of a substrate which is an analyte of interest. Alternatively current max be driven through the electrode to drive reaction of the substrate, for instance to detoxify samples. The redox catalytic domain is suitably derived from a cytochrome P450, and the electron transfer domain may be flavodoxin.

L'invention porte sur une protéine chimère comprenant un domaine catalytique redox provenant d'une source et un domaine de transfert d'électrons provenant d'une source différente. Ladite protéine s'utilise dans un procédé dans lequel le substrat du domaine catalytique redox étant actionné, les électrons sont transférés entre le domaine catalytique redox et le domaine de transfert d'électrons et entre domaine de transfert d'électrons et une électrode. On peut suivre le courant ou le potentiel de l'électrode pour déterminer la présence ou la quantité d'un substrat constituant un analyte d'intérêt. En variante, le courant peut être tiré de l'électrode pour provoquer la réaction du substrat, par exemple pour détoxiquer des échantillons. Le domaine catalytique redox pourra dériver d'un cytochrome P450, et le domaine de transfert d'électrons peut être une flavodoxine.
