DNA is described which codes for chimeric receptors which contain two or more independent polypeptide chains each of which contains an extra cellular ligand association domain attached to a signalling domain through a transmembrane and optionally one or more spacer domains. Each polypeptide chain can be expressed in an effector cell and will remain largely unassociated with the other(s) in the absence of ligand. The presence of ligand induces a stable interaction between the ligand association domains of each chain and facilitates interaction between the intracellular domains leading to a signalling event and activation of the cell. The activated cell may be of use in medicine for example in the treatment of diseases such as cancer.

L'invention concerne de l'ADN codant pour des récepteurs chimères qui contiennent deux ou davantage de chaînes polypeptidiques indépendantes, chacune de celles-ci contenant un domaine d'association de ligand extracellulaire fixé à un domaine de signalisation par une transmembrane, et éventuellement un ou plusieurs domaines d'espaceur. Chaque chaîne polypeptidique peut être exprimée dans une cellule effectrice et reste en grande partie non associée à l'autre/aux autres chaînes en l'absence de ligand. La présence d'un ligand provoque une interaction stable entre les domaines d'association de ligand de chaque chaîne, et facilite une interaction entre les domaines intracellulaires qui aboutit à un événement de signalisation et à une activation de la cellule. La cellule activée peut être utile en médecine par exemple, dans le traitement de maladies telles que le cancer.
