The present invention relates to the purification of large scale quantities of active (infectious) adenovirus and AAV, especially for use in therapeutic applications. In particular, the invention provides improved methods for contacting such viruses with suitable chromatographic materials in a fashion such that any damage to the virus, particularly to surface components thereof, resulting from contact with such chromatographic materials is minimized or eliminated. The result is the ability to rapidly and efficiently purify commercial level quantities of active (infectious) virus suitable for use in therapeutic applications, e.g. gene transfer/therapy procedures.

Purification d'importante quantité d'adénovirus (infectieux) actifs et de virus adéno-associés (AAV), essentiellement pour des applications thérapeutiques. L'invention porte en particulier sur des méthodes améliorées permettant de soumettre de tels virus à des matières chromatographiques appropriées de manière à réduire autant que possible ou éliminer totalement toute altération du virus, notamment des composants de sa surface, par le contact avec de telles matières chromatographiques. Il en résulte une possibilité de purifier rapidement et de manière efficace des quantités d'intérêt commercial de virus actifs (infectieux) se prêtant à des applications thérapeutiques telles que des opérations de transfert de gènes ou de thérapies géniques.
