The present invention relates to chromogenic (colorimetric) in situ hybridization (CISH) and nucleic acid probes useful for in situ hybridization. Specifically, the present invention provides methods, kits, and compositions for performing bright field cancer diagnostics employing chromogenic in situ hybridization (e.g. to detect gene amplifications, gene translocations, and chromosome polysomy). In preferred embodiments, the present invention provides CISH methods, kits and compositions for detecting HER2 gene status.

L'invention concerne l'hybridation chromogène (colorimétrique)in situ (CISH) et des sondes d'acides nucléiques utiles dans l'hybridation in situ. L'invention concerne en particulier des procédés, des kits et des compositions permettant d'effectuer des diagnostics du cancer sur fond clair par l'hybridation chromogène in situ (pour détecter des amplifications géniques, des translocations géniques et une polysomie chromosomique, par exemple). Dans des modes de réalisation préférés, l'invention concerne des procédés de CISH, ainsi que des kits et des compositions permettant de détecter le statut du gène HER2.
