A single incubation CKMB assay is provided which employs an anti-CKB capture antibody bound to magnetic particles and an anti-CKMB tracer antibody labelled with an acridinium ester. The assay achieves a minimal detectable dose of at least 1 ug/1 CKMB for incubations as short as 10 minutes at room temperature, and is not subject to interference due to the presence of macro-CK-1 or high levels of CKMM and CKBB in serum samples. In accordance with certain preferred embodiments of the invention, the anti-CKMB tracer antibody is a mouse monoclonal antibody of the IgG1 type.
