The present invention relates to a method for the clarification of a cell broth containing cells by the following steps: contacting the cell broth with a particulate anion exchange material having a specific density of the particles of between 1.4 and 3, allowing an adequate incubation time, and separating the resulting cell pellet from the supernatant layer. The present inven-tion further relates to a method for the recovery of secreted desired biological substances from a cell broth containing cells pro-ducing the secreted desired biological substance by contacting the cell broth with particulate anion exchange material having a specific density of the particles of between 1.4 and 3 g/ml, allowing an adequate incubation time to result in formation of a cell pellet and a supernatant layer, separating the resulting cell pellet from the supernatant layer and extracting the secreted desired bi-ological substance from the supernatant layer. Optionally, residual biological substance can be further extracted from the resulting cell pellet by one or more washing step.
La présente invention porte sur un procédé pour la clarification d'un bouillon de culture cellulaire contenant des cellules par les étapes suivantes : la mise en contact du bouillon de culture cellulaire avec une matière d'échange d'anions particulaire ayant une densité relative des particules comprise entre 1,4 et 3, l'étape consistant à laisser un temps d'incubation adéquat et la séparation du culot de cellules résultant et de la couche de surnageant. La présente invention porte en outre sur un procédé pour la récupération de substances biologiques sécrétées souhaitées à partir d'un bouillon de culture cellulaire contenant des cellules produisant la substance biologique sécrétée souhaitée par la mise en contact du bouillon de culture cellulaire avec une matière d'échange d'anions particulaire ayant une masse volumique des particules comprise entre 1,4 et 3 g/ml, l'étape consistant à laisser un temps d'incubation adéquat pour entraîner la formation d'un culot de cellules et d'une couche de surnageant, la séparation du culot de cellules ainsi obtenu et de la couche de surnageant et l'extraction de la substance biologique sécrétée souhaitée à partir de la couche de surnageant. Facultativement, la substance biologique résiduelle peut être encore extraite du culot de cellules ainsi obtenu par une ou plusieurs étapes de lavage.
