The present invention is directed to a process for clonally expanding T cells of unknown specificity which comprises co-culturing said T cells of unknown specificity with combinatorial randomized peptide library, Xn, an interleukin, and MHC class II antigen-expressing cells with DNA synthesis being suspended, said cells being derived from the individual where said T cells are obtained, and optionally in the presence of anti-CD29 antibody having an agonist activity. A process for identifying a peptide ligand recognized by said T cell clone of unknown specificity is also provided.
La présente invention porte sur un procédé d'expansion clonale de lymphocytes T dont la spécificité est inconnue. La méthode comprend la coculture desdits lymphocytes T de spécificité inconnue avec une banque de peptides aléatoires combinatoires, Xn, une interleukine et des cellules exprimant des antigènes du CMH de classe II dont la synthèse d'ADN a été arrêtée, lesdites cellules provenant également de la personne chez qui on a prélevé lesdits lymphocytes T, et facultativement, en présence d'anticorps anti-CD29 présentant une activité agoniste. Un procédé permettant d'identifier un peptide ligand reconnu par ledit clone de lymphocyte T de spécificité inconnue est aussi proposé.
