The present invention relates to modified cDNA sequences coding for human Factor X and their derivatives with improved stability and modified activation sequences, recombinant expression vectors containing such cDNA sequences and host cells transformed with such recombinant expression vectors. The invention also relates to recombinant factor X polypeptides and derivatives which do have biological activities of the unmodified wild type protein but with improved stability and processes for the manufacture of such recombinant proteins and their derivatives. The invention also covers a transfer vector for use in human gene therapy, which comprises such modified DNA.

L'invention concerne des séquences d'ADNc modifiées codant le facteur x humain et des dérivés de celles-ci à stabilité améliorée et des séquences d'activation modifiées, des vecteurs d'expression recombinants renfermant de telles séquences d'ADNc et des cellules hôtes transformées au moyen de tels vecteurs d'expression recombinants. L'invention concerne également des polypeptides du facteur x recombinants et des dérivés possédant des activités biologiques de la protéines du type sauvage non modifiée mais avec une stabilité améliorée et des procédés de fabrication de telles protéines recombinantes et des dérivés de celles-ci. L'invention concerne également un vecteur de transfert à utiliser dans la thérapie génique humaine et comprenant un tel ADN modifié.
