A system for releasing a drug specifically in the colon of the gastrointestinal tract, which comprises a drug (b) coated with an organic acid-soluble polymer material (a), and a saccharide (c) which rapidly generates an organic acid by the action of enterobacteria in the lower part of the gastrointestinal tract; a colon-specific drug release oral preparation, which comprises a composition comprising a drug (b) coated with an organic acid-soluble polymer material (a) and a saccharide (c) which rapidly generates an organic acid by the action of enterobacteria in the lower part of the gastrointestinal tract, said composition being coated with an enteric coating polymer material (d). The invention provides a drug release system and a preparation which utilize enterobacteria, which do not form harmful substances due to the release-starting mechanism, show rapid degradation, and have higher colon specificity.
L'invention concerne un système de libération d'un médicament dans le côlon d'un tractus gastro-intestinal. Ledit système comprend un médicament (b) enrobé d'une substance macromoléculaire organique soluble dans l'acide (a), et un saccharide (c) qui génère rapidement un acide organique sous l'action de bactéries intestinales dans un tractus gastro-intestinal inférieur. L'invention porte également sur une préparation de libération de médicament buvable spécifique au côlon comprenant une composition contenant un médicament (b) enrobé d'une substance macromoléculaire organique soluble dans l'acide, et d'un saccharide (c) qui génère rapidement un acide organique sous l'action de bactéries intestinales dans un tractus gastro-intestinal, ladite composition étant elle-même enrobée d'une substance macromoléculaire intestinale (d). Lesdits système et préparation utilisent des bactéries intestinales, ne produisent aucune substance nocive induite par le mécanisme de déclenchement de la libération, et ont une activité élevée spécifique au côlon.
