Methods for making a combinatorial antibody library from human germline segments are provided. Also provided are libraries of nucleic acid molecules compiled from germline segments encoding VL chains and libraries of nucleic acid molecules encoding VH chains, and resulting antibody libraries. The libraries are provided as addressable libraries. Methods for screening antibody libraries against a target protein antigen, and the identified or selected antibodies are provided.

L'invention porte également sur des bibliothèques de molécules d'acide nucléique élaborées à partir de segments de lignée germinale codant pour des chaînes VL, sur des bibliothèques de molécules d'acide nucléique codant pour des chaînes VH, et sur les bibliothèques d'anticorps ainsi obtenues. Les bibliothèques sont élaborées sous forme de bibliothèques adressables. L'invention porte sur des procédés de criblage de bibliothèques d'anticorps contre un antigène protéique cible et sur les anticorps identifiés ou sélectionnés.
