Methods are provided that utilize a naive antibody library for preparing monoclonal antibodies directed against an antigen complex. In particular, methods are provided for preparing antibodies that show specificity for the PSA-ACT complex (prostate-specific antigen complexed to anti-chymotrypsin) as opposed to the free subunits of the complex. Antibodies prepared by these methods are provided.
