Abstract of the Disclosure A composition and method useful in measuring anti- streptolysin O in a blood sample based on the reversible inhibition or inactivation of SO hemolytic activity via pH is herein disclosed. The composition comprises a solution of Streptolysin O having a pH outside the range of about 4.8 to about 9Ø The method includes the step of adjusting the pH of an SO solution to a pH outside the range wherein SO possesses hemolytic activity. More specifically, the method comprises mixing at least one sample of non-hemolyzed blood or serum with at least one predetermined quantity of Streptolysin O (SO) in a non- hemolytic solution having a pH outside the activity range of SO, the SO having a known hemolytic capacity. Then, each solution is incubated at a temperature of about 0-50°C for a time sufficient to allow any ASO in the sample to react with the SO. The hemolytic activity of the SO is restored by adjusting the pH of each in- cubated solution to a range within a pH of about 4.8 to about 9.0, whereby any free SO in excess of the amount required to react with the ASO can lyse blood. Finally, the presence or absence of lysis in each solution is detected.
