There is disclosed a composition and a method for the efficient non-viral delivery of nucleic acids to cells using chitosan. In order to achieve high efficiency of transfection, the composition contains a nucleic acid and a chitosan that has the following physico-chemical properties: a combination of a number-average molecular weight between 8 kDa and 185 kDa and a degree of deacetylation between 72% and 92%. The chitosan molecule can also present additional physiochemical properties such as a block distribution of acetyl groups obtained by a heterogeneous treatment of chitin, and/or a polydispersity index between 1.4 and 7Ø By correctly controlling these parameters, efficient delivery systems may be produced that are effective when optimized for different conditions such as the pH of transfection media and amine-to-phosphate ratio.
L'invention concerne une composition et un procédé utilisant du chitosan pour l'administration non virale efficace d'acides nucléiques à des cellules afin de parvenir à réaliser une efficacité de transfection élevée, ladite composition contenant un acide nucléique et un chitosan qui a les propriétés physicochimiques suivantes : une association d'un poids moléculaire moyen en nombre compris entre 8 kDa et 185 kDa et d'un degré de désacétylation compris entre 72 % et 92 %. La molécule de chitosan peut également présenter des propriétés physicochimiques supplémentaires telles qu'une distribution en blocs des groupes acétyle obtenue par un traitement hétérogène de la chitine et/ou un indice de polydispersité compris entre 1,4 et 7,0. En ajustant correctement ces paramètres, on peut produire des systèmes d'administration efficaces qui sont efficaces lorsqu'ils sont optimisés pour différentes conditions telles que le pH des milieux de transfection et le rapport amine sur phosphate.
