The present application regards compositions containing a compound which modulates the activity of T1-M protein, the use of compounds which modulate the activity of T1-M protein, and an in vitro method for influencing the proliferation and/or differentiation potential of bone cells. Bone differentiation is a complex process in which the proliferation of osteoblasts and the formation of bone matrix play an important role. The signals inducing transition from proliferating osteoblasts to resting bone cells have not been identified up to now. Knowledge of such signals could permit the treatment of osteopetrotic and osteoporotic diseases. According to the invention it has been found that the inhibition of T1-M protein has a proliferation-enhancing effect on osteoblasts whereas its activation stimulates differentiation. Therefore, there are provided compositions which contain compounds capable of modulating the activity of T1-M protein.
La présente demande de brevet concerne des compositions contenant un composé qui module l'activité de la protéine T1-M, l'utilisation des composés qui modulent l'activité de la protéine T1-M et une méthode in vitro pour réguler le potentiel de prolifération et/ou de différenciation des cellules osseuses. La différenciation osseuse est un processus complexe où la prolifération des ostéoblastes et la formation de la matrice osseuse jouent un rôle important. Les signaux déclenchant la transition de la phase de prolifération des ostéoblastes à la phase de quiescence des cellules osseuses n'ont pas encore été identifiés. La connaissance de tels signaux pourrait faciliter le traitement de l'ostéoporose et de l'ostéopétrose. Les inventeurs ont découvert que l'inhibition de la protéine T1-M stimule la prolifération des ostéoblastes alors que son activation stimule la différenciation. Ils présentent donc des compositions qui renferment des composés capables de moduler l'activité de la protéine T1-M.
