Single vector constructs for expression of an immunoglobulin molecule or fragment thereof and methods of making and using the same are described. The vectors comprise a self-processing cleavage sequence between a first and second immunoglobulin coding sequence allowing for expression of a functional antibody molecule using a single promoter. The vector constructs include the coding sequence for a self-processing cleavage site and may further include an additional proteolytic cleavage sequence which provides a means to remove the self processing peptide sequence from an expressed immunoglobulin molecule or fragment thereof. The vector constructs find utility in methods for enhanced production of biologically active immunoglobulins or fragments thereof in vitro or in vivo.
L'invention concerne des vecteurs recombinés uniques permettant l'expression d'une molécule ou d'un fragment d'immunoglobuline, et des procédés de préparation et d'utilisation de ces vecteurs. Ces vecteurs contiennent une séquence de clivage autocatalytique placée entre une première et une deuxième séquence de codage d'immunoglobuline, permettant l'expression d'une molécule d'anticorps fonctionnelle à l'aide d'un seul promoteur. Ces vecteurs recombinés contiennent une séquence codant pour un site de clivage autocatalytique, et peuvent contenir en outre une séquence de clivage protéolytique additionnelle qui sert à éliminer la séquence peptidique autocatalytique de la molécule d'immunoglobuline exprimée ou du fragment de celle-ci. Ces vecteurs recombinés sont utiles dans des procédés permettant une production accrue d'immunoglobulines biologiquement actives ou de fragments de celles-ci in vitro ou in vivo.
