The present invention relates to the microbial immunogens engineered to bear .alpha.-gal epitope(s) for induction of potent humoral and cellular immune responses when administered to subjects having anti-Gal antibodies. In one embodiment, the present invention provides compositions and methods for propagating influenza virus in human, ape, Old World monkey or bird cells that have been engineered to express an .alpha.l,3galactosyltransferase (.alpha. l,3GT) gene to produce virions bearing hemagglutinin molecules containing .alpha.-gal epitopes, to increase the immunogenicity of the influenza virus. In another embodiment, the present invention provides fusion proteins between influenza virus hemagglutinin and a microbial peptide or protein of interest, and enzymatic processing of this fusion protein to carry .alpha.-gal epitopes, to increase the immunogenicity of the microbial peptide or protein of interest.
La présente invention concerne les immunogènes microbiens produits par ingénierie pour porter un ou plusieurs épitopes .alpha.-gal pour l'induction de réponses immunitaires humorales et cellulaires puissantes lorsqu'ils sont administrés à des sujets ayant des anticorps anti-Gal. Dans un mode de réalisation, la présente invention propose des compositions et des procédés de propagation du virus influenza dans des cellules d'humain, de grand singe, de singe de l'Ancien Monde ou d'oiseau qui ont été produites par ingénierie pour exprimer un gène .alpha.l,3galactosyltransférase (.alpha.l,3GT) pour produire des molécules d'hémagglutinine portant des virions contenant des épitopes .alpha.-gal, pour augmenter l'immunogénicité du virus influenza. Dans un autre mode de réalisation, la présente invention propose des protéines de fusion entre l'hémagglutinine du virus influenza et un peptide microbien ou une protéine microbienne d'intérêt, et le traitement enzymatique de cette protéine de fusion pour porter les épitopes .alpha.-gal, afin d'augmenter l'immunogénicité du peptide microbien ou de la protéine microbienne d'intérêt.
