The invention provides compositions and methods for inhibiting Chk1 and/or Chk2 kinases. Also provided are compositions and methods for inhibiting G2 cell arrest checkpoint, particularly in mammalian, e.g., human, cells. The compositions and methods of the invention are also used to treat disorders of cell growth, such as cancer. In particular, the invention provides methods for selectively sensitizing G1 checkpoint impaired cancer cells to DNA damaging agents and treatments. Also provided are methods for screening for compounds able to interact with, e.g., inhibit, enzymes involved in the G2 cell cycle arrest checkpoint, such as Chk1 and/or Chk2/Cds1 kinase.
L'invention concerne des compositions et des méthodes d'inhibition de kinases Chk1 et/ou Chk2. Elle concerne également des compositions et des méthodes d'inhibition du point de contrôle de l'arrêt cellulaire G2, notamment dans des cellules mammaliennes, par exemple, humaines. Lesdites compositions et méthodes de cette invention sont utilisées pour traiter des troubles de croissance cellulaire, tels que le cancer. Notamment, la présente invention concerne des méthodes de sensibilisation sélective des cellules cancéreuses altérées à point de contrôle G1 aux traitements et agents endommageant l'ADN. Elle a aussi trait à des méthodes de criblage de composés capables d'interagir avec des enzymes impliqués dans le point de contrôle de l'arrêt du cycle cellulaire G2, tel que la kinase Chk1 et/ou Chk2/Cds1, lesdits composés étant capables par exemple d'inhiber ces enzymes.
