Disclosed are novel proteins, referred to as megakaryocyte growth and development factors (MGDFs; also generally referred to as Mpl ligands), that have a biological activity of stimulating the growth of megakaryocytes and augmenting the differentiation or maturation of megakaryocytes, ultimately to result in the production of platelets. MGDF derivatives comprising MGDF molecules attached to water- soluble polymers, such as polyethylene glycol, are also disclosed, along with methods for their preparation. Also disclosed are processes for obtaining the MGDFs in homogeneous form from natural sources and producing them by recombinant genetic engineering techniques from mammals, including humans.

Nouvelles protéines, appelées de facteurs de croissance et de développement des mégacaryocytes (MGDF; appelées également ligands myéloprolifératifs), ayant pour effet biologique de stimuler la croissance des mégacaryocytes et d'augmenter leur différentiation ou leur maturation, pour entraîner finalement la production de plaquettes. L'invention porte également sur des dérivés MGDF comprenant des molécules MGDF liées à des polymères solubles dans l'eau, comme le polyéthylène-glycol, et sur leurs méthodes de préparation. Elles portent également sur les procédés d'obtention des MGDF sous forme homogène à partir de sources naturelles et sur leur production par des techniques génétiques de recombinaison à partir de mammifères, dont l'être humain.
