The invention provides a method for reducing or inhibiting net beta-amyloid peptide production and amyloid plaque formation associated with Alzheimer's Disease by administering to a subject an amount of a netrin-1 polypeptide, wherein the netrin-1 polypeptide comprises an amino acid sequence that a) binds to a naturally occurring APP protein and b) inhibits A.beta. peptide production. A netrin-1 polypeptide has an amino acid sequence sufficient for specific binding of the netrin-1 polypeptide to the APP protein. A netrin-1 polypeptide can mimic netrin-1 -mediated signal transduction by altering the localization, protein-protein binding and/or enzymatic activity of an intracellular protein involved in an APP signal pathway. The invention also provides a method for reducing or inhibiting amyloid plaque formation associated with Alzheimer's Disease by administering to a subject an amount of a netrin-1 therapeutic, wherein the netrin-1 therapeutic comprises an amino acid sequence that a) binds to a naturally occurring APP protein and b) inhibits A.beta.peptide production.
L'invention concerne une méthode permettant de réduire ou d'inhiber la production nette de peptides bêta-amyloïdes et de la formation de plaques amyloïdes associées à la maladie d'Alzheimer, par l'administration à un sujet d'une dose d'un polypeptide nétrine-1, lequel comprend une séquence d'acides aminés qui a) se lie à une protéine APP naturelle, et b) inhibe la production de peptides A.beta.. Le polypeptide nétrine-1 possède une séquence d'acides aminés suffisante pour lui permettre de se lier spécifiquement avec la protéine APP. Le polypeptide nétrine-1 peut imiter la transduction du signal activée par la nétrine 1 en modifiant la localisation, la liaison protéine-protéine et/ou l'activité enzymatique d'une protéine intracellulaire intervenant dans la voie de signalisation des APP. L'invention concerne en outre une méthode de réduction ou d'inhibition de la formation de plaques amyloïdes associées à la maladie d'Alzheimer, consistant à administrer à un sujet une dose d'un agent thérapeutique à base de nétrine-1, cet agent thérapeutique à base de nétrine-1 contenant une séquence d'acides aminés qui a) se lie à une protéine APP naturelle et b) inhibe la production de peptides A.beta..
