The present invention concerns methods and compositions for identifying human targeting peptides sequences. In preferred embodiments, a primary phage library is administered to a human in vivoand phage bearing targeting peptides are collected from selected organs, tissues or cells types. In more preferred embodiments, collected phage are amplified, pooled and administered to a second human for another round of selection. The peptides are of use for targeted delivery of therapeutic agents, including gene therapy vectors. Such targeted delivery may be used for detection, diagnosis or treatment of human diseases. In certain embodiments, the peptide may be attached to an imaging agent and administered to a human to obtain an image or to diagnose a disease state. Also disclosed are a large number of targeting peptide sequences and consensus motifs that are selective for human organs or tissues, obtained by the methods of the present invention.

La présente invention concerne des procédés et des compositions permettant d'identifier des séquences de peptides ciblants humains. Selon des modes de réalisation préférés, on administre à l'homme une échantillothèque de phages primaires, puis on recueille des peptides ciblants porteurs de phages dans des organes, tissus ou types de cellules choisis. Selon des modes de réalisation encore plus préférés, on prend les phages recueillis et on les amplifie, on les regroupe, et on les administre à un second humain pour un autre cycle de sélection. Ces peptides conviennent à l'administration ciblée d'agents thérapeutiques, y-compris des vecteurs de thérapie génique. Une telle administration ciblée convient à la détection, au diagnostic ou au traitement de maladies humaines. Selon certains modes de réalisation, le peptide peut être attaché à un agent de contraste d'imagerie et administré à un homme pour obtenir une image ou pour diagnostiquer un état pathologique. L'invention concerne également un grand nombre de séquences de peptides ciblants et de motifs de consensus sélectifs d'organes ou de tissus humains, et que l'on obtient selon les procédés de l'invention.
