Compositions and methods effective in inhibiting abnormal or undesirable cell proliferation, particularly endothelial cell proliferation and angiogenesis related to neovascularization and tumor growth are provided. The compositions comprise naturally occurring, or synthetic proteins, peptides, or protein fragments capable of binding to components of the blood coagulation pathway. The compositions may be administered using a pharmaceutically acceptable carrier. The methods involve administering to a human or animal the composition described herein in a dosage sufficient to inhibit cell proliferation, particularly endothelial cell proliferation. The methods are useful for treating diseases and processes mediated by undesired and uncontrolled cell proliferation, such as cancer, particularly by inhibiting angiogenesis. Administration of the composition to a human or animal having prevascularized metastasized tumors is useful for preventing the growth or expansion of such tumors.
L'invention concerne des compositions et des méthodes pouvant inhiber efficacement une prolifération cellulaire anormale ou indésirable, notamment la prolifération de cellules endothéliales et l'angiogenèse liée à la néovascularisation et à la croissance tumorale. Ces compositions contiennent des protéines naturelles ou synthétiques, des peptides, ou des fragments de protéine pouvant se lier à des éléments de la voie de coagulation sanguine. Les compositions de l'invention peuvent être administrées au moyen d'un transporteur pharmaceutiquement acceptable. Les méthodes de l'invention consistent à administrer la composition à un être humain ou à un animal, selon une posologie propre à inhiber la prolifération cellulaire, notamment la prolifération de cellules endothéliales. Ces méthodes sont utiles pour le traitement de maladies et de processus induits par une prolifération cellulaire indésirée et incontrôlée, telle que le cancer, notamment par inhibition de l'angiogenèse. L'administration de la composition de l'invention à un être humain ou à un animal présentant des tumeurs prévascularisées ou métastasées peut empêcher efficacement la croissance ou la progression de ces tumeurs.
