Five protein families cooperate to form the enediyne ring structure, also referred to as the warhead structure, that characterizes enediyne compounds. This divisional application is directed to one of the five protein families, namely the polyketide synthase protein family (PKSE). The invention of the divisional application relates to members of the PKSE family and to nucleic acids encoding members of the PKSE family. The invention of this divisional application also relates to expression vectors and host cells containing the nucleic acids, methods of making a polypeptide of the PKSE family, and methods of identifying enediyne biosynthetic gene clusters and enediyne producing organisms using the nucleic acids and polypeptides of the invention. The invention further relates to use of the nucleic acids and polypeptides to design new enediyne compounds.
