Methods of making and compositions that stimulate a protective immune response in a subject include a portion of a protein from a naturally occurring viral hemagglutinin, wherein the portion includes at least a portion of a globular head, includes at least a portion of at least one secondary structure that causes the globular head to essentially retain its tertiary structure, and lacks a membrane fusion domain, a transmembrane domain and a cytoplasmic domain. Compositions comprise a flagellin component or a Toll-like Receptor agonist component that is at least a portion of a flagellin or a Toll-like Receptor agonist, wherein the flagellin component or Toll-like Receptor agonist component includes at least one cysteine residue and whereby the flagellin component or Toll-like Receptor agonist component activates a Toll- like Receptor 5 or Toll-like Receptor. Compositions comprise a flagellin component that is at least a portion of a flagellin, wherein at least one lysine of the flagellin component has been substituted with at least one arginine, serine and histidine, whereby the flagellin component activates Toll- like Receptor 5. Compositions can further include an antigen, such as an influenza antigen, a flavivirus antigen, a pathogen-related antigen, a bacterial capsular antigen and a carrier protein. The compositions are used to stimulate an immune response and a protective immune response in a subject.
La présente invention concerne des procédés de fabrication et des compositions qui stimulent une réponse immunitaire protectrice chez un sujet comprenant une partie d'une protéine dérivée d'une hémagglutinine virale naturelle, ladite partie comprenant au moins une partie d'une tête globulaire, contenant au moins une partie d'au moins une structure secondaire qui permet à la tête globulaire de retenir sensiblement sa structure tertiaire, et est déficiente en un domaine de fusion membranaire, en un domaine transmembranaire, et en un domaine cytoplasmique. L'invention concerne également des compositions comprenant un constituant de flagelline ou un agoniste du récepteur de type Toll qui est au moins une partie d'une flagelline ou d'un agoniste du récepteur de type Toll, ledit constituant de flagelline comportant au moins un résidu de cystéine et le constituant de flagelline ou l'agoniste du récepteur de type Toll assurant l'activation d'un récepteur 5 de type Toll ou d'un récepteur de type Toll. L'invention concerne en outre des compositions comprenant un constituant de flagelline qui est au moins une partie d'une flagelline, ladite au moins une lysine du constituant de flagelline ayant été substituée avec au moins une arginine, une sérine et une histidine, ledit constituant de flagelline assurant l'activation du récepteur 5 de type Toll. Les composition peuvent également comprendre un antigène, tel qu'un antigène de la grippe, un antigène de flavivirus,un antigène lié à un agent pathogène, un antigène capsulaire bactérienne et une protéine de support. Les compositions sont utilisées pour stimuler une réponse immunitaire protectrice chez un sujet.
